Laboratory Parts:
- Part 1: Students prepare the gels and DNA samples (this usually takes 30-40 minutes)
- Part 2: Students run their gels (while gels are running, have students work on their "Concept Questions" handout)
- Part 3: Students calculate RFLP lengths using banding patterns and size approximation*
*Important Note: Teachers have a choice in how students will further analyze their gel data in Lesson 3-3. You may choose to have students only do the first 7 steps of Part 3 so that the size approximation option can take place in Lesson 3-3 or so that you can instead use the semi-log plot option also outlined in Lesson 3-3.
Laboratory Objectives:
- To understand the physical properties of DNA molecules that allows their separation by agarose gel electrophoresis and apply this technique to an engaging scenario.
- To understand the relevance of this technique to scientific research and global issues.
- To provide students with further experience with the techniques of electrophoresis, micropipetting, and sample preparation.

- Gel Electrophoresis Kit, including freezer box with DNA samples (provided by SEP)
- Microwave ovens or hot plates
- Balances
- Deionized or distilled water (recommended, but not essential)
